Resumo: | Lectinas são proteínas ou glicoproteínas de origem não imune que apresentam pelo menos um domínio não catalítico que reconhecem e liga-se de maneira reversível às moléculas específicas de carboidratos e possuem habilidade de aglutinação de células animais e/ou células vegetais e ação de precipitar glicoconjugados. São amplamente distribuídas na natureza, estando presente em sua maioria na família Leguminosae, no qual uma variedade de lectinas deste grupo já foram isoladas e caracterizadas. Este estudo possibilitou id entificar a presença de uma lectina em sementes de B. pulchella pertencente à família Leguminosae, subfamília Caesalpinioideae. Das extrações obtidas em diferentes valores de pH, a lectina apresentou a melhor solução tampão de extração protéica foi o TRIS HCl 0,1M pH 7,6, pois no extrato bruto preparado observou-se uma maior atividade hemaglutinante. Nos ensaios de hemaglutinação, observou-se que a lectina aglutinou fortemente eritrócitos de coelho tratados com papaína e tripsina após uma 1 h de incubação a 37 ºC. Após 12 h de incubação, detectouse que a lectina aglutinou fracamente eritrócitos humano do tipo B e O. Nos ensaios de inibição da atividade hemaglutinante, a lectina mostrou especificidade por Galactose, β -lactose e α-lactose, apresentando maior especificidade de ligaçã o por β-lactose. |
Descrição: | Lectins are proteins or glycoproteins of non-immune origin that have at least one non catalytic domain that recognize and bind reversibly to specific carbohydrate molecules and have agglutination ability of animal cells and/or plant cells and the action of precipitating glycoconjugates. They are widely distributed in nature, being mostly present in the Leguminosae family, in which a variety of lectins from this group have already been isolated and characterized. This study allowed to identify the presence of a lectin in seeds of B. pulchella belonging to the family Leguminosae, subfamily Caesalpinioideae. From th e extractions obtained at different pH values, the lectin presented the best solution of the protein extraction buffer was TRIS HCl 0.1M pH 7.6, because in the prepared crude extract a greater hemagglutinating activity was observed. In the haemagglutinatio n assays, it was observed that the lectin was strongly agglutinated papain and trypsin -treated rabbit erythrocytes after a 1 h incubation at 37 °C. After 12 h incubation, the lectin was detected to poorly agglutinate human B and O type erythrocytes. In the hemagglutinin inhibition assays, lectin showed specificity for Galactose, β-lactose and α-lactose, exhibiting increased binding specificity for β-lactose. |
